Malaria mitochondrial diagnosis: challenges and pitfalls

چکیده

Background High-copy genomic sequences could be used as PCR targets for the detection of Plasmodium infections, providing increased sensitivity over single- or low-copy genes. Mitochondrial genomes malaria parasites are present in multiple copies a single mitochondrion, and each parasite has many mitochondria. Here, we describe development seven species-specific qPCR assays diagnosis vivax falciparum , targeting coding non-coding mitochondrial regions. Methods The optimization protocols involved gradient annealing temperatures concentrations primers probes, well inclusion additives/enhancers [e.g., dimethyl sulfoxide (DMSO), glycerol, bovine serum albumin (BSA)] to improve specificity amplification. Results Non-specific amplification other species human was observed different levels all assays. Regardless late Cq values most non-specific amplifications, application cutoff value did not completely exclude false-positive amplification, compromising also Conclusions Therefore, although have higher sensitivity, they frequently lose due their high sequence conservation. A screening evaluate cross-reaction between malaria-free samples must performed

Challenges and pitfalls in the diagnosis of hyperprolactinemia.

The definition of the etiology of hyperprolactinemia often represents a great challenge and an accurate diagnosis is paramount before treatment. Although prolactin levels > 200-250 ng/mL are highly suggestive of prolactinomas, they can occasionally be found in other conditions. Moreover, as much as 25% of patients with microprolactinomas may present prolactin levels < 100 ng/mL, which are found...

Mitochondrial disorders: Challenges in diagnosis & treatment

Mitochondrial dysfunctions are known to be responsible for a number of heterogenous clinical presentations with multi-systemic involvement. Impaired oxidative phosphorylation leading to a decrease in cellular energy (ATP) production is the most important cause underlying these disorders. Despite significant progress made in the field of mitochondrial medicine during the last two decades, the mo...

Laboratory diagnosis methods of malaria infection

Background and Aim: To manage and control malaria - one of the most important infectious diseases in tropical and subtropical regions – a quick and accurate malaria diagnosis method is necessary as a high priority. For this purpose, several methods have been developed for the diagnosis of malaria, and currently, the examining the peripheral blood slide with an optical microscope as a gold stand...
